The interaction of polypeptide components of human high density serum lipoprotein with detergents.

چکیده
The cationic detergent tetradecyltrimethylammonium ion interacts with four binding sites on the surface of AI polypeptide from human serum high density lipoprotein and 10 sites on the AI1 polypeptide with association constants of 2 X lo4 and 1 x lo4 liters per mole, respectively. The sites on AI appear to be primarily hydrophobic in that the anionic detergent sodium dodecyl sulfate binds to the same number of sites with the same association constant as the cationic amphiphile. Cooperative binding and a concomitant protein conformational change at higher free concentrations of ligand ( >10e4 M) are observed for both AI and AI1 with these two detergents as well as with deoxycholate. It is suggested that this altered protein state may differ from those observed for water-soluble proteins in high concentrations of tetradecyltrimethylammonium ion or dodecyl sulfate.

Study of Serum Paraoxonase and High Density Lipoprotein Fractions in Diabetes
Objective: Significant alteration in lipid profile and antioxidant system occurs in response to diabetes mellitus (DM). Paraoxonase (PON) is a family of three enzymes PON1, PON2 and PON3 associated with high density lipoprotein (HDL). The HDL in human plasma consists of two main sub-fractions HDL2C and HDL3C. We studied the HDL subclasses and HDL associated enzyme paraoxonase with respect to di...
متن کاملThe Effect of Eight Weeks of High-intensity Interval Training on Lipoprotein-associated Phospholipase A2 and Lipid Profile in a Male Rat Model of Type 2 Diabetes
Introduction: Type 2 diabetes (T2D) causes hyperglycemia, hyperinsulinemia, and dyslipidemia, which are all risk factors for atherosclerosis. Lipoprotein-associated phospholipase A2 (Lp-PLA2) has been recognized as an indicator of atherosclerosis due to its role in vessel inflammation. This study aimed to investigate the effect of high-intensity interval training (HIIT) on serum levels of Lp-PL...
متن کاملSerum Lipid Profile Alterations in Acute Leukemia Before and After Chemotherapy
Background: Serum lipids abnormalities have been observed in various forms of cancers including acute leukemia. Investigators report decreased total cholesterol and HDL, and elevated triglyceride in leukemic patients. Limited studies had been performed to discover the correlation between abnormal lipids profile and disease activity in leukemic patients in Iran. This study was done to evaluate t...
متن کاملEffects of Urtica dioica hydro-alcoholic extract on blood serum glucose and lipid profiles of female Wistar rats with long-term estrogen deficiency
In this study, the effects of Urtica dioica hydro-alcoholic extract were investigated on the blood glucose and lipid profiles of female ovariectomized and non-ovariectomized rats. In total, 32 adult female rats were divided into four groups (eight each) including control and ovariectomy groups as well as non-ovariectomy and ovariectomy groups treated with 200 mg kg-1 of ...
متن کاملConformational stability of the polypeptide components of human high density serum lipoprotein.
An investigation of the denaturation of the major polypeptides from human serum high density lipoprotein by guanidine hydrochloride reveals that the lipid-free, water-soluble states are minimally stable relative to the random coil states. These findings are in direct contrast to the resistance of intrinsic membrane proteins (in the absence of ligand) to complete unfolding by the same denaturant...
